It's more of a running gag making fun of difficulties Mizuki-san's had in meeting up with Takeshi-san to hand over doujinshi to him, plus riffing off the odd behavior of the osa-yukkuri. This time, Takeshi-san apparently snuck up on Mizuki-san (unwittingly, no doubt) and startled him just by being there.

you can almost hear the "Mon-mon!"
