A lit cigarette leaves trails of tobacco behind while the DS Stylus does not. Cigarettes are all organic materials wrapped in paper while the DS stylus is made of plain hard plastic.

If a lit cigarette is used to play the DS, the touch screen may be dirty and, if you're unfortunate, break.
