It's a little hard to see, so flats are vectored in using Photoshop, and the rendering is mostly the brush tool in SAI, with Blending at 0 and Dilution and Persistence pretty low. In advanced options, he also has Edge Hardness at 100.

Looks alive in this picture. The snake's head is raised up and its tongue is sticking out.
