My guess is he probably did and the result of that relationship is who Priscilla is, and maybe Yorshka.

What she's calling him in Japanese script? Because "Big brother" or "Elder brother" means much more in Japanese than in English, and its meaning and weight don't translate well.

Maybe she has a big crush on him?

Because Gwyn was fighting the dragons and the Nameless King became a traitor by siding with the dragons against Gwyn. Many kings/gods don't take kindly to their kin turning traitor, I'd imagine. Plus the world of Dark Souls doesn't seem very big into the whole "forgive and forget" thing.

I can buy Priscilla being a possible grandchild of Gwyn but isn't Yorshka the youngest child of Gwyn's children? Really that would make Yorshka and Priscilla aunt and niece.

There isn't a Japanese script. Bloodborne is the only Soulsborne game with one.
